Hit the spooky ones first then let the drug get them down before shooting the rest. Pen size really depends on the temperment of the deer (how close you can get to make a shot without spooking them) your proficiency with a dart or blow gun and the terrain anf fauna that would prevent you from getting a good shot. If the pen is laid out correctly, a 2 acre pen may suffice.

You may already be doin this but I always reccomend nightime darting.It takes a little getting used to but is much easier to get good shots and deer dont run around nearly as much therefore sressing themselves less.It also helps because deer that are still up when pulling out tranked deer will be much less likely to bounce off the walls at night.
